Happy to share our latest review @CellCellPress with @ruferbus and Wolfgang Baumeister about high resolution imaging methods (#cryoET #SuperResolution) to investigate neurotoxic protein aggregates in #neurodegeneration. ➡️doi.org/10.1016/j.tcb.2020.0…

Get ready for #SPACEtomo 🚀🌌 - fully automated parallel tomography on #cryoFIB lamellae. 🤖🔬❄️🦠 Another amazing development by @EisFabian 🙂👍
biorxiv.org/content/10.1101/…
We developed "DeepFIBing" - a new strategy to create #cryoFIB-lamellae for #cryoET in any depth of very large cells - this also works for tissue. If you want to know more, have a look into our publication in CELL with @jentoft_ida and @SchuhLab. #teamtomo bit.ly/3u3Azp6
Advantages of DeepFIBing:
- A simple method, which allows to create lamellae in any depth inside large specimens.
- Ice crystal contamination becomes irrelevant: a new contamination-free surface is created.
- Does not require high-pressure freezing or lift-out equipment.

What is the optimal thickness of a Cryo-FIB lamellae? We quantify the tradeoff between specimen thickness and FIB damage by subtomogram averaging. There is no need to go ultra thin. Work by M Tuijtel, B Turonova @jpkreysing @sonjawelsch @HummerLab now online. @MPIbp #teamtomo

What an honor to receive a Consolidator grant from @ERC_Research !! We are excited to dive into nervous tissues using #cryoET in our #cryoNERVE project 🤩
Thanks to everyone who made this possible!
It was quite a ride to pick the LS1 @ERC_Research #ERCCoG 2022 grantees from such a great group of people so HUGE congratulations to: @ruferbus , @DoraTang5 , @Hashemyaser4 , @LaineElodie , @LandauMeytal , @TzviyaZBM , @YiliangDing and Stefano Pagliara (@LSI_Exeter) !!!! 🥳

Do you wonder about the future potential of #cryoET?
Will #AlphaFold replace experimental methods in structural biology or complement them?
In our new review with Wolfgang Baumeister we discuss these questions in the Special Issue “AlphaFold” @JMolBiol.

25 years ago I was measuring whales stranded dead along Sardinian coast, or releasing back to sea those found alive. Now our cryo-EM paper on the cetacean morbillivirus nucleoprotein-RNA complex is out on @JStructBiol sciencedirect.com/science/ar…

We have updated our joint preprint with @ruferbus on #lysosome defects caused by protein aggregation, and added new data on the mechanism:
Amyloid-like aggregates cause lysosomal defects in neurons via gain-of-function toxicity biorxiv.org/content/10.1101/…

Been working with @phospho animation on amyloid aggregation. 1st effort is pretty stunning, I think! This is an in vitro, wild-type hIAPP fibril from @R_U_Gallardo @RadfordLab lovely paper in @NatureSMB @Astbury_BSL @AstburyCentre #LoveTheWigglyBits

In practice, is #cryoET limited to cells in culture? In this manuscript we show an easy way to image native tissues with molecular detail. 1/5

Are you interested in #cryo-ET of native tissues?
In our new pre-print we propose an easy applicable approach using plunge freezing and #cryoFIB to investigate vitrified tissue by #cryoET. With Jose Pastor-Pareja & @ruferbus. #cryoET_histology #cryoEM

#cryoFIB lamellae come often with disturbing preparation artifacts. We developed an algorithm, that computationally removes these and facilitates the interpretation of the rich context of these lamella overviews: „Lamella in silico Clearing“ (LisC)

Many physiological & pathological processes depend on the complex association of cells and extracellular structures in the tissue context which cannot be recapitulated in culture. Visualized here by #cryoET: a peripheral nerve with axons, perineurial glia & basement membrane

We demonstrate the applicability of this approach on nervous tissue, muscle tissue and epithelial tissue with molecular resolution. #cryoET_histology
