Postdoc @bara_anissa presenting at #WCBR2022 on the effects of prenatal #cannabis exposure on developmental reprogramming in the placenta! @MountSinaiPsych @winter_brain
Finally, STRING PPIs identified other epigenetic regulators in the Kmt2a complex that were altered in our datasets, emphasizing the profound effects of PTE on epigenetic regulation.
Next, we used CRISPR to upregulate Kmt2a in neural progenitor cells, and RNA-seq showed perturbations *again* to synaptic plasticity-related genes, and gene-gene correlations similar to our in vivo data!
Conducting ChIP-seq with H3K4me3 (Kmt2a target) in the NAc revealed a surprising overlap of genes with H3K4me3 enrichment and *reduced* mRNA expression!
We conducted RNA-seq to elucidate the effects of PTE on the NAc transcriptome and identified overlapping DEGs between DESeq2 and limma related to synaptic and neuronal functioning.
Measuring the expression of numerous genes in the NAc coding for epigenetic regulators revealed upregulation of Kmt2a (gene and protein levels) 🧬; knockdown of Kmt2a in the NAc normalized motivation for food.
Starting with behavior, we observed that PTE increased motivation for food reward and induced depression-like behavior, but had opposite effects on hedonia based on prior stress experience.
Challenging human cannabis study, we thank the brave mothers for their participation in these discoveries.
"Maternal cannabis use is associated with suppression of immune gene networks in placenta and increased anxiety phenotypes in offspring."
