1/n. Final version now online @MolMetab! Especially for labs newer to metabolomics, we hope this clarifies the need to freeze tissues immediately after loss of perfusion to preserve the in vivo metabolism. sciencedirect.com/science/ar…
Ever wondered how quickly mouse tissues need to be frozen to preserve the in vivo metabolome? Immediately after disrupted perfusion. We show here that this applies to the broad metabolome, between-genotype differences, and 13C-isotopologue distributions. biorxiv.org/content/10.1101/…

Hello everyone! I am helping to organize a one-day symposium on fluorescent metabolite biosensors and their use in metabolism research. It will be May 12-13 and associated with the Cold Spring Harbor Laboratory “Mechanisms of Metabolic Signaling” Symposium. Our field needs to be able to visualize metabolism with spatial and temporal resolution and this symposium will explore what to do and how to do it, with experts in metabolism and biosensor development and use. Please join us! meetings.cshl.edu/meetings.a… -JR
How do cells respond to physiologically high O2 levels? In research just published in Science Advances (science.org/doi/10.1126/scia…), we investigating respiratory epithelia since their contact with air exposes them to some of the highest O2 levels in the body.
In work led by @B581434 (Bo Ram Kim) and co-corresponding with Mike Welsh, we show that basal airway stem cell mitochondria signal high O2 levels via mitochondrial citrate export.
High O2-induced mitochondrial citrate export leads to selective histone acetylation and differentiation into ciliated airway cells, which are critical for clearing mucus and pathogens from the lungs. Our findings may inform mechanisms to enhance lung airway repair after injury or treat other conditions where the number of ciliated cells is insufficient.

Have you ever wondered how mitochondrial and cytosolic pathways for de novo lipogenesis (DNL) interact?
We did!
Published Online today (cell.com/cell-metabolism/ful…) and led by first-author @adam_rauckhorst, we traced 13C-labeled lactate/pyruvate, acetate, the ketone acetoacetate, and leucine in several mouse models of disrupted hepatic mitochondrial and cytosolic acetyl-CoA generating pathways.
Surprisingly, deleting the mitochondria citrate carrier (CiC) increased DNL from both exogeneous and leucine-generated acetoacetate. Overall, our findings delineate a mitochondrial-cytosolic DNL substrate supply network that may guide more accurate therapeutic development for MASLD and T2D.
Many thanks to our expert collaborators: No X Ryan Sheldon, now leading metabolomics at the @VAInstitute, who helped start the project and develop the necessary mass spec methods with @adam_rauckhorst; for @StanislawDeja and @Isotopomer for deuterium tracing by NMR and careful contibutions to quantitatively framing our mass spec data; and Tom Vallim of @TarlingVallim for guidance with in vivo gene deletion by AAV CRISPRs.
Please also see the exciting co-published manuscript from @WellenLab and Christian Metallo Labs (cell.com/cell-metabolism/ful…) showing that bempadoic acid does more than just inhibit ACLY!
