Really encouraging data. And trop2 is ubiquitous except for NEC and sarcomatoid. Exciting data for challenge!
SAC-TMT + pembro shows 68% response rates in metastastic bladder cancer #ESMO2026 . Trofuse-55 explores this combination in bladder sparing approaches in a randomized phase 3. The triplet with EVP seems a justified research randomised question. Triplet data from yesterday showed 81% RRs. @OncoAlert

Are our pathology guidelines keeping pace with reality? 📖 A thought-provoking proposal to make diagnostic criteria practical, accessible and globally applicable in this excellent paper by the authors - Philippe Benjamin Stephenson, Olaleke Folaranmi, José Jiménez Heffernan, and Sanjay Mukhopadhyay, published in the BMJ Journal of Clinical Pathology. Three powerful messages: 🔹 Make criteria feasible across resource settings 🔹 Focus on what truly impacts diagnosis and treatment 🔹 Guideline panels should include community pathologists and represent the global community of pathologists The best guidelines should not just define what is ideal—they should help every pathologist make the best possible diagnosis with the resources available. Sending federal election monitors to New Haven is more about intimidating voters than protecting election integrity. There is no evidence of fraud or illegality—not a scintilla—that justifies this overreaching by a DOJ that should be pursuing real wrongdoing.

#Weekend_Review : Venetoclax for Rx of #ALL Venetoclax is increasingly being explored across B-ALL and T-ALL, from frontline therapy to relapsed/refractory disease. What have we learned so far, and where might it fit? 1/10 First, why venetoclax? BCL-2 is a key anti-apoptotic protein. Venetoclax selectively inhibits BCL-2, lowering the threshold for mitochondrial apoptosis. But the important concept is: BCL-2 expression ≠ BCL-2 dependence. That distinction may help explain both sensitivity and resistance.

🧬🔬 MOSAIC is out in Genome Medicine! The Multi-Omics Spatial Atlas in Cancer: >2,700 tumor samples across ≥10 tumor types, generated in standardized labs at 5 founding partners (CHUV, Charité, UK Erlangen, Gustave Roussy, Pitt) together with Owkin (@OwkinScience) 📦 6 modalities from ONE FFPE block:
• H&E whole slide images
• Visium CytAssist spatial transcriptomics
• snRNA-seq (Chromium Flex)
• Bulk RNA-seq
• Whole exome sequencing
• Curated clinical data 🔓 MOSAIC Window, the first open release: 60 patients with bladder cancer, ovarian cancer, GBM, DLBCL and mesothelioma, available via EGA. Key findings:
🧩 Most tumors harbor ≥2 genomically, spatially and transcriptionally distinct malignant subsets (pan-cancer median 3, GBM median 6)
🫧 Bladder cancer shows the highest inter-patient heterogeneity of all indications
🔗 Tumor cell signaling tracks with its neighborhood: TGF-β ↔ fibroblasts, VEGF and JAK-STAT ↔ macrophages
🧬 Copy number profiles do not always dictate transcriptomic state, pointing to passenger alterations 4 case studies with a therapeutic angle:
🎯 Mesothelioma: an inflamed, MET-active niche with high anti-PD-1 signature next to an immune-cold niche in the same tumor
🎯 Bladder: squamous/keratinizing vs. proliferative subsets independent of CN background; CEACAM5/6 confined to nest cores, relevant for ADC targeting
🎯 Ovarian: HRD signature restricted to one subset, suggesting heterogeneous PARPi sensitivity 💡 Bulk profiling averages out exactly the niches that may decide treatment response. Proud that Erlangen Pathology is a founding partner. Thanks to all patients, the Owkin team and every co-author 🙏. This is just the first step on a great journey with MOSAIC #PathTwitter #SpatialTranscriptomics #SingleCell #Multiomics #DigitalPathology #AIinPathology #PrecisionOncology #TumorHeterogeneity #BladderCancer #OpenData 📄 doi.org/10.1186/s13073-026-0…

Some of my colleagues think things will just “blow over” and “it’s not that bad”. This @nytimes article starts to reveal the deep wounds this govt has (self)inflicted on the research engines that have driven US innovation. All I can say is VOTE next week! nytimes.com/2026/10/31/clima…

A truly disturbing report on the administration's effort to politicize and dismantle the scientific infrastructure in the US. This should be concerning to all who care about the health of their loved ones, all businesses concerned with the welfare of their employees and their future products, and all those who care about the long term economic and military strength of America. Disappointing catch to the announcement about interceptors going to Ukraine—delivery in a few years. Facing a hideously harsh winter & searing hardship from Russian bombs, Ukraine urgently needs air defense.
A strong decision by the U.S. State Department – authorizing Ukraine’s purchase of Patriot interceptors to counter Russian ballistic missiles. Five hundred PAC-3 interceptors is a substantial number, and very soon – this winter – they COULD make for a highly effective defense package. However, it has been stipulated that deliveries may begin ONLY IN A FEW YEARS. We are asking our American partners to find a solution for the time when there is a real danger and a real need to use PAC-3 interceptors. Right now, the Russians are attacking our energy infrastructure every single day. In just these past two weeks of October, there have already been more than 30 Russian strikes on critical parts of our power grid. It is right now, even before the beginning of winter, that Putin has started carrying out his operation to attack the systems that sustain life in our cities and communities. The targets are electricity, heating, water supplies, bridges, and life in our cities in general. We did ask our American partners to provide Ukraine with a winter support package – at a rate of 100 Patriot interceptors per month. Five hundred missiles would be enough to protect our skies over the coming months – precisely when it is so important for Ukraine to be in a strong position in negotiations and for Russia NOT to succeed in breaking life in Ukrainian cities and communities. The United States, our European partners, and other partners around the world have sufficient stocks of PAC-3 interceptors in their warehouses. Providing them to Ukraine right now – in the winter of 2026–2027 – and replenishing our partners’ arsenals LATER, in THE FOLLOWING YEARS, through this production already contracted and paid for by Ukraine and our partners would be the right approach. And it is entirely feasible. We are grateful to President Trump. Importantly, we recently discussed PAC-3 interceptors with Secretary of State Marco Rubio. Ukraine is grateful to the U.S. State Department and the U.S. Administration overall for granting this authorization. But we are asking for decisions that can guarantee the protection of lives not the day after tomorrow or sometime later, but at the very moment the threat is there – when Russia is killing and destroying. I thank everyone who stands with Ukraine! state.gov/releases/bureau-of…

Mass photometry uses light to weigh individual molecules. I think it’s really compelling and useful. If you wanted to weigh a protein, for example, you shine light at it, and electrons in the protein respond by scattering some of that light. The amplitude of the scattered light wave is approximately proportional to molecular mass, meaning that larger proteins produce a "stronger" signal than smaller proteins. Special microscopes can detect these tiny, tiny signals by combining light scattered by a protein with light reflected from the glass slide beneath it. The two light waves interfere, producing a small change in brightness. Researchers calibrate those measurements using proteins with known masses. Now, usually, mass photometry weighs a molecule as it falls on top of glass. But for this paper, they describe a method to keep weighing the same molecule and watch its mass change as other molecules attach and detach, in real-time. Basically, they attached individual proteins to small patches of a lipid membrane, so each protein could move a little bit, but couldn’t drift out of view. They then measured the mass of that protein 497 times PER SECOND over the span of an entire hour. Every time the protein binds to anything (like an antibody, or its receptor, or a small molecule), the binding event shows up as a little spike in the data. So with the SARS-CoV-2 spike protein, for example, they could follow its mass rising and falling as the ACE2 receptor attached and detached. They measured how long the two stayed bound together, also: 64 seconds on average. They also watched antibodies attach one after another, producing up to three distinct increases in mass, matching the spike’s three binding sites. Really beautiful biophysics.

Very happy to share newest online publication in JAMA Oncology @JAMAOnc, "FGFR Alterations in Bladder Cancer- From Molecular Pathogenesis to Precision Oncology," written by Dr. Katrina Collins @katcollmd In this review, we examine how FGFR biology is reshaping the molecular classification and precision management of patients with bladder cancer. We highlight the central role of FGFR3 alterations and the fast-growing range of molecular biomarker testing at every stage of disease. We discuss comprehensive genomic profiling, liquid biopsy (including urine-based molecular testing), and emerging molecular residual disease (MRD) assessment. We also cover how each is used in diagnosis, treatment selection, surveillance, and disease monitoring. On the therapeutic front, erdafitinib has established FGFR-directed precision therapy in advanced urothelial carcinoma. Combination approaches, intravesical FGFR-directed treatments, and next-generation FGFR3-selective inhibitors are extending this strategy into new clinical settings. Emerging technologies, including AI-assisted histopathology and integrated multi-omic biomarkers, may further refine patient selection. Read the full article: doi.org/10.1001/jamaoncol.20…
