5/ To understand the non-cell autonomous effects of ERK dynamics, we used co-cultured monolayer system. In co-cultures, cells with sustained ERK activity trigger waves of ERK signaling in neighboring cells, while cells with pulsatile ERK activity do not. (Gif in first tweet 👆)
4/ Pulsatile ERK signaling increased #Proliferation as expected, but sustained ERK activation causes i) nuclear ERK kinase translocation, ii) increased cell migration, and surprisingly, iii) #CellCycle arrest.
3/ We made an MCF10A cell line expressing both the ERK KTR #biosensor to track ERK activity, and an ERK localization sensor. Using a TET-inducible system, we can observe differences in ERK dynamics following #Oncogene induction by #LiveImaging.
