[Log #ΞL-026] Compound: $RAP (Rapamycin) Subject: Gold Standard of Longevity Observation: While new compounds and cocktails are emerging and may surpass Rapamycin in certain models, the community and leading experts agree: Rapamycin remains the gold standard of longevity. As Krister Kauppi stated, “the road to bypass Rapamycin is long.” Rapamycin was the first to take longevity research from caloric restriction into the era of true gerotherapeutics. Whether it is: – $RAPTOR (SMER28 + Rapamycin) launched by @eperlste – $RULI (Rapamycin + Lithium + Urolithin A) launched by @Rulicompound – $RAPGU (Rapamycin + Urolithin A + Ginsenoside Rh2) launched by @drug_devv …the common denominator is always Rapamycin. Interpretation: Rapamycin is to longevity what cement is to the foundation of a building: indispensable, enduring, and the basis upon which future progress will be built. Next Steps: – Ongoing $RAP experiments on flies and mice – Continued validation and benchmarking against emerging compounds

🧾 Log #ΞL–Final] - Closure Announcement & RAP Transition Observation: After more than a year of work, passion, and relentless effort, the Eterna Labs team has made the difficult decision to shut down the DAO. Since our launch on July 23rd, we have poured our energy, time, and personal funds into building something meaningful for the longevity ecosystem. Despite our best efforts, and after deep internal discussions, we’ve reached the conclusion that the DAO model is no longer the right vehicle to carry this mission forward. As part of this closure, 100% of the assets held in the AUM will be converted in SOL and fully returned to all investors. We have never sold a single token from the treasury, and we have personally invested over $20,000 of our own funds to buy back supply and support the floor throughout the project. Interpretation: Even though Eterna Labs is ending, the story of Rapamycin the molecule that carried the heart of our vision does not end here. Rapamycin has always been bigger than us. Bigger than a single team. Bigger than a DAO. And that’s why we are deeply grateful to announce that Rapamycin Longevity Lab (@OMIPALRLL) will officially take over the remaining supply and lead the community takeover of $RAP. RLL has been a cornerstone of the longevity movement, and their dedication to Rapamycin research is unmatched. With them, $RAP will not only live on, it will continue to grow, evolve, and reach the heights it deserves. This transition ensures continuity, integrity, and long-term focus. The molecule is in the best hands possible. Implication: The closure of Eterna Labs marks the end of a chapter, but not the end of the mission. By handing $RAP to RLL, we’re ensuring: A smooth, transparent transition for all holders A clear scientific vision led by the people most committed to Rapamycin A real chance for $RAP to reach its full potential with a team fully aligned and deeply experienced The preservation of everything we built together We may be stepping back, but the research, the progress, and the spirit of acceleration live on. Status: The DAO is officially closing. All AUM funds will be returned to holders. The remaining $RAP supply will be transferred for a full CTO by RLL. Eterna Labs will no longer operate, but the Rapamycin project continues under a stronger, more focused leadership. From the bottom of our hearts, thank you. Thank you for believing in us, for building with us, for caring about the science, and for pushing this vision forward. It’s with a heavy heart that we close this chapter, but we know the best part of the story is still ahead. Rapamycin is going to the top, and you’re in very good hands.

🧾 [Log #ΞL-060] — Rapamycin Enters the Hair Healthspan Race Observation: Fresh data dropped on Pump Science: Doxycycline delivered an extraordinary +80% median lifespan increase in C. elegans, making $DOCS the current King of the Pill in worms. Alongside this, the combinations Rapamycin + Doxycycline ( $RAPADOCS ) and Omipalisib + Doxycycline also completed their worm runs. While some speculated that Doxycycline’s effects may come from altering bacterial food (calorie restriction mimicry), multiple labs, using both live and dead bacteria, now report lifespan extension that cannot be explained by CR alone. This confirms a true mechanistic signal, not an artifact. But the real story is what comes next. Interpretation: This is not just about validating worm results. This is about translating longevity mechanisms into human-relevant outcomes. And the most immediately impactful frontier is hair healthspan: - reducing androgenic hair loss, - preventing or reversing gray hair, - and promoting hair rejuvenation. Both Rapamycin and Doxycycline already have promising preclinical and early human data in hair regeneration pathways, through mTOR modulation, inflammation control, mitochondrial stress signaling, and stem-cell niche preservation. This is why a new startup has been launched, in collaboration with Rapamycin Longevity Lab, to push these findings into real consumer biotech products, not just scientific notes. In this context, Rapamycin remains the anchor compound: the most studied life-extension molecule ever discovered, now entering its most promising cosmetic application yet. Implication: - Rapamycin ( $RAP ), Doxycycline ( $DOCS ) and RAP + DOCS ( $RAPADOCS ) tokens will now co-fund applied hair research, including mouse studies, formulation testing, safety validations, and early human pilot trials. - Once commercialization begins, a share of product revenue will be used to buy back these tokens, creating a full loop where basic science → product → market → research reinforcement. Parallel to worm data, fly experiments for Doxycycline, RAPADOCS, and Omipalisib+Doxycycline are next, with strong expectations that Doxycycline may dominate in flies as well. Rapamycin stands at the center of this ecosystem, in longevity, in immune modulation, in aging biology, and now in hair rejuvenation biotech. Status: The startup is now active. Research direction is set: hair loss, gray hair, rejuvenation. Token-funded experiments continue. Mouse studies and fly assays are being prepared. One molecule led the longevity revolution. Now it may lead the aesthetic longevity revolution. Rapamycin is entering a new chapter, from lifespan to hair healthspan.

🧾 [Log #ΞL-059] — Rapamycin Restores Ovarian Function in Endometriosis Models Observation: A new study published in Molecular Medicine Reports (Wu et al., 2025) demonstrates that short-term rapamycin treatment can restore ovarian function and fertility potential in mice with endometriosis. Researchers observed that endometriosis, which accelerates ovarian aging and damages follicular quality, was partially reversed following rapamycin administration. Key outcomes included: - Decreased ovarian senescence markers (p16, p21, γH2AX). - Increased antioxidant enzymes (SOD, GPx) and mitochondrial function. - Restoration of follicle structure, granulosa cell health, and oocyte quality. - Normalized hormone-receptor and PPARα–IGFBP2 signaling pathways. Interpretation: This study reinforces the role of mTOR inhibition as a regulator of female reproductive longevity. Rapamycin not only slows cellular aging but also appears to rejuvenate ovarian tissue, improving both structural integrity and hormonal balance. By targeting oxidative stress and senescence, rapamycin could represent a therapeutic path to extend fertility windows and mitigate age-related ovarian decline, two hallmarks of reproductive aging. Implication: While still pre-clinical, these results align with the growing body of evidence that rapamycin’s longevity effects extend to specific organ systems, here, the reproductive axis. For longevity research initiatives, it opens a high-impact avenue: fertility preservation through senescence modulation.

[Log #ΞL-057] — Correction Notice: Article Reference Update Observation: In our previous post regarding thymic regeneration, we mistakenly quoted the wrong study link. Correction: The intended reference was the recent Rapamycin News coverage of the paper “Thymic regeneration in female mice following short-term rapamycin administration.” nitter.gq/RapaNews/status/198475… Note: Our apologies for the earlier mislink, scientific accuracy remains our priority. We appreciate the community’s vigilance in helping maintain the integrity of shared data. Status: Rapamycin continues to reveal systemic rejuvenation potential across multiple aging pathways.
🧾 [Log -056] — Thymic Regeneration in Female Mice Following Short-Term Rapamycin Observation: In a new study, female mice (BALB/c) treated with low-dose Rapamycin (1 mg/kg/day for 3 days, i.p.) showed dramatic thymus involution followed by regeneration. Within 14 days after treatment cessation, the thymus mass rose to ~130% of baseline. Key findings included: • ~46% organ shrinkage at Day 3 (weight loss, disrupted medullary/subcapsular TECs, shorter thymocyte telomeres) • By Day 14: regained architecture, increased double-positive (CD4+CD8+) and CD8+ single positive T-cells, telomere lengthening, and upregulated Foxn1/Klf6, Sirt3/Sirt6. • Additional interventions (IL-7, Metformin) modified specific biomarkers (telomere length, mitochondrial health) but did not accelerate volumetric thymic recovery. Interpretation: This reveals a biphasic response to mTOR-inhibition: initial stress (involution), followed by regenerative rebound. The data suggest Rapamycin may act as a “reset” signal for thymic renewal, rejuvenating immune architecture rather than merely slowing decline. The effect, observed in young females, hints at systemic immune regeneration potential despite initial suppression. Implication: For longevity research, this opens a new domain: immune-system rejuvenation via mTOR pathways. • For Eterna Labs and its partners Rapamycin Longevity Lab (RLL) & AgelessRx: this mechanistic foothold deepens the case for Rapamycin’s role beyond lifespan, now immune‐span. • Future protocols can integrate thymic and immune biomarkers (telomeres, T-cell subsets, Foxn1/Klf6 expression) in human trials. • This supports selecting immune functionality as a measurable endpoint in decentralized human protocols: tracking protection against infection, immune resilience, and cellular renewal. Status: Though pre-clinical and limited to young female mice, the study charts a novel regenerative route. Eterna Labs will incorporate these metrics into upcoming Rapamycin human-data collection frameworks, continuing to build the world’s most comprehensive dataset on mTOR & lifelong resilience. From lifespan to immunity, Rapamycin’s story continues to expand. Reference: Thymic regeneration in female mice following short-term rapamycin administration. Li J et al., 2025 (Eur. J. Pharmacol.). 🧾 [Log #ΞL-055] — Short-Term Rapamycin Treatment Improves Embryo Quality, Pregnancy & Live Births Observation: A recent publication in Cell Reports Medicine (via Rapamycin News) presents data showing that short-term rapamycin treatment in aged human reproductive tissues led to improved embryo quality, higher pregnancy rates and successful live births. Key mechanistic observations include: - Aged oocytes/cumulus cells show elevated ribosomal gene activity, reduced expression of meiosis/actin/cohesin genes, and signs of lysosomal dysfunction. rapamycin.news+1 - Rapamycin treatment suppressed translation load (reduced ribosomal-mRNA output, lower p-S6 and p-4E-BP1 signalling), improved proteostasis (reduced misfolded protein aggregates), and shifted cells toward a more youthful translational profile. - Functionally, in a cohort of women with repeated IVF failures, rapamycin was associated with high-quality embryos and successful pregnancies-live births, suggesting translational relevance for human fertility. Interpretation: This study underscores that rapamycin’s impact may extend beyond lifespan and health-span into reproductive span, directly influencing oocyte competence, embryo viability and live birth outcomes in older individuals. By targeting ribosome biogenesis and translation burden (a novel mechanism in aging), rapamycin appears to restore proteome balance in aged reproductive tissues, consistent with fundamental geroscience concepts. Within the broader ecosystem of Eterna Labs, Rapamycin Longevity Lab (RLL) and AgelessRx, this finding adds a powerful new data point: functional human outcomes (fertility, embryo quality, live births) rather than only biomarker or model-organism endpoints. Implication: For the longevity & fertility research intersection, this paper provides: - A mechanistic target: translation load/ribosome dysregulation → cellular aging → tissue dysfunction. - A human clinical outcome: improved embryo quality + live births, which are strong functional endpoints. - A new use-case for Rapamycin: extending reproductive window/competence, aligning with Eterna Labs’ mission to expand health-span in the broadest sense. Status: While still early (pilot data, niche cohort), the study represents a key turning point: rapamycin now shows human reproductive benefit, not just theoretical or pre-clinical. The human database expands, the translational gap narrows. We are entering a new phase: where longevity, reproduction and health-span converge. From cellular translation to human generation, rapamycin’s impact spans life itself.

🧾 [Log #ΞL-052] — Low-Dose Rapamycin in ME/CFS: Pilot Evidence Observation: A decentralized, uncontrolled study involving 86 patients with Myalgic Encephalomyelitis/Chronic Fatigue Syndrome (ME/CFS) assessed the effects of low-dose Rapamycin (6 mg/week) over 30–90 days. Key outcomes: - 72.5% of 40-patient subset showed notable improvements in post-exertional malaise (PEM), fatigue (MFI), and quality of life (SF-36). - Autophagy biomarkers shifted: ↑ BECLIN-1, ↓ pSer258-ATG13; correlated with symptom improvements. - Treatment was well tolerated: no serious adverse events; lab shifts (WBC ↓, LDL/HbA1c ↑) remained within normal ranges. Interpretation: This study adds to the body of evidence that rapamycin via mTOR inhibition and autophagy activation, may reduce core symptoms of ME/CFS (fatigue, PEM) in a real-world, decentralized framework. The correlation between biomarker change (autophagy marker modulation) and clinical outcomes strengthens the mechanistic plausibility of effect. The decentralized design aligns with DeSci ideals: open participation, real-world settings, broad data capture. Implication: For Eterna Labs this pilot underscores three key opportunities: - Expand human self-experimentation protocols under regulated, data-rich frameworks. - Integrate autophagy/biomarker read-outs into our data architecture to identify responders and optimize dosing curves. - Leverage decentralized clinical platforms (e.g., AgelessRx) to scale access and data capture for low-dose rapamycin interventions. Status: While the study is uncontrolled and has inherent limitations (non-placebo design, ME/CFS-specific cohort), it sets a benchmark: rapamycin is progressing beyond lifespan models into symptom-relief and mechanistic human work. Eterna Labs will integrate these findings into our research roadmap, targeting controlled trials, broader populations, and standardized biomarker-driven protocols. The evidence for rapamycin is shifting fast, not just living longer, but living better. Source: pubmed.ncbi.nlm.nih.gov/4050…

🧾 [Log #ΞL-051] — Rapamycin in Motion Observation: The Rapamycin ($RAP) compound has now completed its first flies movement trials on @pumpdotscience, marking another decisive step in the experimental chain. Across 45 independent fly races, Rapamycin-treated flies won 44 times against controls, demonstrating a consistent enhancement in vitality and mobility. Average speed in the final experiment reached 3.46 px/s versus 0.14 px/s for controls, a ~25× difference. Distance covered mirrored this effect, with Rapamycin flies maintaining higher travel ranges throughout most trials before age-related decline appeared late in the series. In fly models, locomotor activity is one of the most reliable healthspan biomarkers, reflecting neuromuscular integrity, mitochondrial performance, and systemic energy metabolism. High movement = high health. Interpretation: These results reinforce Rapamycin’s profile as a healthspan enhancer, not only extending lifespan, but maintaining physiological function over time. Movement in CANTON-S (flies) is directly correlated with survival and metabolic robustness, making this performance gain a key signal of mTOR pathway optimization. As seen in the data: - RAP consistently outperformed controls in both speed and distance metrics, especially during midlife stages. - The steady decline in control group activity contrasts with the sustained locomotor resilience of RAP-treated flies. Eterna Labs and Rapamycin Longevity Lab will continue correlating these movement dynamics with lifespan curves to determine the translation potential for mammalian and human healthspan markers. Implication: With worm, fly, and soon mice experiments active, Rapamycin now demonstrates cross-species robustness in performance enhancement. The next step: bridging these biological signals into human data collection through the ongoing collaboration between Eterna Labs, Rapamycin Longevity Lab, and AgelessRx. Motion is a mirror of vitality. Rapamycin doesn’t just extend life, it sustains movement. Link: pump.science/experiments/RAP…
