Great to see independent confirmation: (R)-lipoic acid inhibits HDACs, but (S)-lipoic acid doesn‘t. @ParisRWatson from @ChristiansonLab delivers beautiful structural basis for the enantioselective binding to HDAC6 - congrats!
Our first preprint! Congrats to @ParisRWatson and @jubsnjubs for showing how the well-known cofactor of central carbon #metabolism, reduced lipoic acid, binds with high nanomolar affinity to HDAC6: biorxiv.org/content/10.1101/…

Together with @medardlab, we prepared the attached letter describing our concerns with using ricolinostat as an HDAC6 inhibitor. Thoughts/feedback are welcome.
We plan to submit this to @NatureCancer for consideration as a "matters arising" letter.
Very disappointed to see this new paper in @NatureCancer on a clinical trial with “HDAC6 inhibitor” ricolinostat. The paper fails to mention three recent reports demonstrating that this drug exhibits pervasive off-target toxicity.
