Great question! In our cohort, we saw rapid evolution (< 6 mo) of PPM1D ± TP53-mutated clones following radioligand therapy, CAR-T, and PARPi-based therapy. The tempo and clinical trajectory, however, appeared to vary with the concurrent evolution/acquisition of additional somatic or chromosomal hits. Link to paper 📝 doi.org/10.1182/bloodadvance…

Sad that the @myESMO embargo was broken... It is difficult to compare results between an adjuvant study and a neoadjuvant study (ie, S1801) in any cancer, including #melanoma. The patient populations were different and the primary endpoint (RFS vs EFS) was of necessity different for technical reasons that as @adamfeuerstein points out don't translate well to this platform. Until tomorrow's presentation we won't have all the data necessary to even try and compare. It is also important to note that NO drugs are technically @FDA approved for neoadjuvant treatment for melanoma. It's a moot point as several studies including S1801 and NADINA have made neoadjuvant therapy standard of care in some situations. I think it's great that the study (that I participated in) appears to have hit its primary endpoint, which typically ultimately translates into FDA approval. Tomorrow (1030 ET) will be interesting...

Alzheimer’s is one of the most complex, heartbreaking challenges in modern healthcare. And if we’re ever going to make real progress against a disease this devastating, we need two things: unprecedented data scale and computing power. When you bring together @bmsnews drug development expertise, @pathos agentic AI, and @TempusAI data infrastructure, you create something rare: a real opportunity to uncover novel insights, fix flawed trial designs, and actually accelerate precision treatments for patients who can't afford to wait.
Today we announced a strategic collaboration with @bmsnews to support their oncology drug portfolio. We also announced a strategic collaboration with Bristol Myers Squibb and @pathos to advance drug discovery in Alzheimer’s disease. Collectively, this makes BMS one of our largest data and modeling customers. Through this collaboration, BMS researchers will be able to train advanced AI models using de-identified data to surface breakthroughs and increase the probability of success when developing therapies for both cancer patients and those suffering from Alzheimer’s disease. 🇨🇳 TQB2102 (biparatopic HER2 TOPO1 ADC) also passes the truck test in mostly HR+/HER2-low MBC. PFS 16.4 vs 5.5 mo, HR 0.29, p<0.01. Unclear whether the biparatopic design adds anything. The HER2 TOPO1 field is getting quite crowded with excellent agents. #ESMOAmbassadors #ESMO26

Excellent presentation by Jens presenting results from KN866 trial that did not meet its primary endpoint in contrast with NIAGARA. Peri-op EV-pembro is the SOC in MIBC, awaiting VOLGA trial tomorrow! @myESMO #ESMO26 #ESMOAmbassadors @OncoAlert @Uromigos @shilpaonc @AndreaNecchi
Full house for Bladder/RCC updates first with #KN866 #ESMO26 Gem/cis in MIBC + placebo or pembro (➖) study & less relevance in the era of EVP SOC Vs similar trial NIAGARA was ➕ pCR 44.4% vs 35.8% EFS & OS not significant We wait #VOLGA tomorrow for more in this space

When the immune system is weakened by cancer treatment — especially by a stem cell or bone marrow transplant (BMT) for blood cancer — common viruses like herpes simplex virus (HSV) can become a serious problem. For someone who is immunocompromised, an HSV infection can mean dealing with large, painful sores that resist the usual therapies and ultimately lead to more critical medical conditions. The good news: The @FDA recently approved #pritelivir (Hovilpri) for HSV infection in immunocompromised patients, including those who are undergoing cancer treatment. The study that resulted in the drug’s approval was co-led by Dr. @GenPapaMD, Clinical Director for Infectious Diseases at MSK, and Dr. June Lee, an infectious disease specialist at MSK, among others. I really admire figures that can describe a concept better than long manuscripts. This image is from an excellent paper in Nature by Vivek Subbiah about the future of evidence-based medicine and the design of clinical trials, all inspired by the rise of artificial intelligence. This is the era of the next generation of 'deep' medicine. "Current paradigms must be continuously challenged by emerging technology and by all stakeholders (the new generations of scientists, physicians, the pharma industry, regulatory authorities and, most importantly, patients). Disruptive innovation should lead to every clinical site being a research site, with all necessary quality checks and research as part of the standard of care. My personal view is that every major disease needs a ‘Moonshot’ program and every rare disease should have an ‘Operation Warp Speed’—both with clearly identified, sustainable goals to improve population health and address equity, diversity and global access to therapies. Methodological advances and future AI-based analyses of all data will provide deep evidence to realize the goal of personalized medicine— that is, to offer the right treatment to the right patient at the right time."
