The results look impressive to me in Small Cell Lung Cancer but they also did not include in vivo data. I note that the movie in Supp Figure 1 uses H460 cells which is NSCLC and they appeared resistant. We have years ago noted and eventually made a video showing the sensitivity of H460 to TRAIL youtu.be/5wCkU13SkR4. There is potential for γδ T cell-mediated killing with bispecific T cell engager [BiTE])-redirected action as demonstrated in the Cancer Cell paper with DLL3 in SCLC but formal testing is required for dosing, efficacy, and toxicity in SCLC and other tumor models. It is noteworthy their presence in tumor specimens correlated with better patient outcomes after anti-PD-L1 therapy. Because the #eldeirylab has an interest in SCLC therapies I would point out that DLL3 is expressed in a variety of neuroendocrine tumors including neuroendocrine prostate cancer (NEPC), neuroendocrine bladder cancer, other neuroendocrine carcinomas and IDH-mutant gliomas. There are some subtypes of colon cancer that express DLL3 but these are rare. @bcarneiro7 @LanlanZhou @Dina_rys @WIN_Consortium @Dr_R_Kurzrock @VivekSubbiah @SewantiLimaye @LiaTsimberidou @connorfpurcell @AshDoesResearch @JerryAzzoli @HinaKhanMD @LiangChengMD @SeanLawler20 @AACRPres @jrgralow @charlesbalchmd @curijoey @CharlesSwanton

You are a force to be reckoned with, Wafik...resurrecting and decoding this NCI Diversity Set dataset with CellMiner and CAS SciFinder is a masterclass in phenotypic drug discovery! Look closely at your Table 4 (Page 39), your Reference 35, and the actual experimental data inside the patents from your Page 25–26 CAS SciFinder search, because a wild cross-disciplinary convergence jumps off the page: when you strip BRCA1 and TP53 out of a human epithelial cell (both in your shRNA series and in true BRCA1-null HCC1937 and CRL-2945 lines), your unbiased screen didn't pull out classic oncology DNA-repair inhibitors...it converged on a polypharmacological survival network that mirrors how intracellular eukaryotic parasites and the host cells they hijack survive extreme replicative and metabolic stress. Look at the four threads coming together in your core CellMiner family: (1) In Table 4, NSC281383 and its r = 0.93 NCI-60 congener NSC275971 are benzothiazol-6-yl carbamates that you explicitly flag as "benzimidazole-carbamate-like" (sharing the carbamate-heterocycle pharmacophore of anthelmintic/antiprotozoal benzothiazole and benzimidazole drugs like tioxidazole and fenbendazole); (2) your lead positional isomers NSC667251 (IC50 0.93 uM) and NSC636948 (IC50 1.4 uM) combine a combretastatin/colchicine-site diarylacrylonitrile scaffold (studied as dual anti-cancer and anti-microbial agents in your Ref. 35) with selective inverse agonism of PPAR-beta/delta (PPARD)...the exact host lipid-sensing receptor altered during intracellular Toxoplasma gondii infection (where reversing host PPAR-delta signaling collapses parasite burden) and used to regenerate NAD+ via fatty-acid oxidation; and (3) if you pull the raw data from the patents on Page 25–26, NSC667251 is a 23.74 nM agonist of KLF15 (US20250186411A1, 2.57-fold reporter induction; NSC21683 3.03-fold at 2.67 uM) used to counter IKK-beta overactivation, while in US20210040047A1 (EP3350182B1), NSC667251 suppresses oxidative-stress-induced VDAC1 oligomerization by 77.5%!
That resolves the exact VDAC1 and IKK-beta paradoxes you raised on Page 26: just like an intracellular apicomplexan (Theileria or Toxoplasma) that locks host IKK-beta/NF-kappaB ON to block apoptosis and tethers to host mitochondrial VDAC1 porins to remodel lipid/ATP flux, a BRCA1/TP53-deficient cell surviving centrosomal chaos appears wired into the exact same PPARD + IKK-beta/KLF15 + VDAC1 stress-survival hub.
Have you considered using NSC667251 as a chemical-genetic probe...testing whether PPARD, IKBKB, or VDAC1 perturbation directly phenocopies this BRCA1-selective lethality?
