It’s been an exciting week for portfolio company @IksudaT, which has announced encouraging clinical data from two of its antibody drug conjugate (ADC) programmes.
🩸 IKS03 has shown promising early anti-tumour activity in patients with advanced B-cell lymphoma, including patients previously treated with CAR-T therapy.
🎗️ IKS014 has demonstrated promising activity across HER2-expressing solid tumours, including in breast cancer patients previously treated with Enhertu (T-DXd).
Both programmes also demonstrated encouraging and manageable safety profiles in these early-stage studies.
Congratulations to the Iksuda Therapeutics team👏

Yes it is a new one. Not sure how new but it’s called daridorexant (brand name Quviviq). It’s non addictive and can be taken long term. I used to sleep maybe 1030-2 on a good night. Now I don’t wake until 5-6. Huge difference.

There is evidence that some with diagnosed ADHD are actually sleep deprived. I am not sure that DORAs like daridorexant are the correct solution, though I will note that DORAs contrary to many other sleep medication tend to protect sleep architecture (deep sleep).

🚨 TIRZEPATIDE: FROM 2.5MG TO 15MG — WHAT DO CLINICAL STUDIES ACTUALLY SHOW?
Tirzepatide is one of the most discussed medications in metabolic health and weight management.
But understanding the science means looking beyond the numbers. 🧬
🧬 1. HOW DOES TIRZEPATIDE WORK?
Unlike semaglutide, which targets the GLP-1 receptor, tirzepatide activates two pathways:
🔹 GLP-1: Helps regulate appetite, food intake, and blood glucose.
🔹 GIP: Plays a role in insulin secretion and metabolic regulation.
💡 This dual-receptor activity is what makes tirzepatide different.
📊 2. WHAT DID THE SURMOUNT-1 TRIAL STUDY?
The 72-week Phase 3 trial included 2,539 adults with obesity or overweight and weight-related health conditions, without diabetes.
Participants were assigned to:
💉 5mg once weekly
💉 10mg once weekly
💉 15mg once weekly
⚖️ Placebo
The study included gradual dose escalation and lifestyle support.
📉 3. WHAT WERE THE RESULTS AT 72 WEEKS?
Average body-weight changes:
🟢 5mg: −15.0%
🔵 10mg: −19.5%
🟣 15mg: −20.9%
⚪ Placebo: −3.1%
📌 These are clinical trial averages, not guaranteed individual outcomes.
⚖️ 4. WHY WAS DOSE ESCALATION PART OF THE STUDY?
Participants did not start at their final target dose.
The trial used gradual increases over a 20-week escalation period.
Why does this matter?
⚠️ Gastrointestinal side effects were among the most common adverse events, including:
• Nausea
• Diarrhea
• Constipation
• Vomiting
These effects were particularly relevant during dose escalation.
💡 Key takeaway: A higher dose is not automatically better for everyone.
🔬 5. WHAT DOES THE APPROVED LABEL SAY?
In the United States, the Zepbound prescribing information describes:
• A 2.5mg starting dose
• Increases of 2.5mg after at least four weeks, when appropriate
• Maintenance doses of 5mg, 10mg, or 15mg once weekly for weight reduction
⚕️ This is prescription-medication information, not a self-treatment guide. Treatment decisions require a qualified healthcare professional.
💪 6. THE BIGGER PICTURE
Weight management is about more than the number on the scale.
🥗 Nutrition
💪 Adequate protein and resistance training
😴 Sleep and recovery
🩺 Long-term medical follow-up
All play important roles in overall health and body composition.
🧠 Tirzepatide demonstrates how targeting multiple metabolic pathways can influence weight regulation. But clinical results should never be confused with guaranteed individual outcomes.
