🧬🔥 What if omega-3 fatty acids could reprogram brown adipose tissue to release factors that kill cancer cells?
A new study in Nutrition & Metabolism reveals that DHA-enriched omega-3 supplementation remodels adipose endocrine function and enhances brown adipose tissue (BAT) secretome-mediated melanoma cell death.
🔬 1. DHA reshapes systemic inflammation
Female C57BL/6 mice received DHA-enriched omega-3 (DHA = 5:1; 1 g/kg/day) for 35 days.
Supplementation reduced:
• IL-1β: approximately threefold
• IL-6 and TNF-α: approximately twofold
• Oxidant-sensitive fluorescence in peritoneal lavage cells
Body weight remained unchanged.
🧬 2. Adipose remodeling occurs without weight loss
DHA-enriched supplementation reduced:
• Gonadal WAT mass: ~50%
• Subcutaneous WAT mass: ~40%
• Gonadal adipocyte size
BAT mass remained unchanged, but BAT TGF-β protein levels declined significantly.
Classical thermogenic and lipolytic markers were largely unchanged.
⚡ 3. BAT secretome acquires enhanced antitumor activity
Researchers collected conditioned media (CM) from BAT and subcutaneous WAT and exposed B16-F10 melanoma cells to these secretomes.
BAT-CM from DHA-supplemented mice produced:
• Increased Annexin V-positive melanoma cells
• Greater total melanoma cell death
• Approximately 40% reduction in MTT-assessed metabolic viability
Importantly, BAT-CM preserved viability in nonmalignant NIH/3T3 fibroblasts under the tested conditions.
🔥 4. The effect is depot-specific
Although both BAT- and WAT-derived CM reduced melanoma viability, DHA supplementation selectively enhanced BAT-CM cytotoxicity.
BAT-CM also exhibited increased triglyceride concentrations.
The specific cytotoxic lipid or secreted factor remains unidentified.
🔋 5. Adipose secretomes alter tumor inflammatory signaling
Conditioned media from DHA-supplemented animals reduced melanoma-associated IL-6 production.
TGF-β levels also shifted toward unstimulated control values.
These observations suggest that adipose endocrine remodeling may influence tumor-associated inflammatory programs.
🎯 Therapeutic implications
Dietary DHA → Adipose endocrine remodeling → Altered BAT secretome → Enhanced melanoma cell death in vitro
This proposed adipose–tumor communication pathway offers a potential direction for metabolic cancer research.
However, the study did not demonstrate tumor suppression in vivo, identify the responsible secreted mediators, or establish clinical anticancer efficacy.
💡 Key takeaway: Brown adipose tissue may influence cancer biology through its secretome independently of measurable changes in BAT mass or classical thermogenic markers. DHA supplementation provides a potential nutritional approach to modifying this endocrine communication.

Pup calls can wake sleeping mouse mothers, and Tang, Wu and colleagues identified one circuit involved in that response. Neurons in the temporal association and ectorhinal cortices that project to the hypothalamic preoptic area respond more strongly to pup calls in mothers than in virgin females, a difference linked to higher expression of the dopamine D1 receptor. During NREM sleep, the auditory thalamus continues to transmit information about the pups’ ultrasonic calls to this cortical pathway. Manipulating the circuit changed how often mothers woke to pup calls, while waking to nonsocial sounds depended less on it.
I’d love to know whether fathers and other caregivers develop comparable changes through caregiving.

🧬🔥 What if sympathetic nerves burn fat not only through norepinephrine—but by remodeling the extracellular matrix surrounding adipocytes?
A new study in Nature Metabolism identifies GUSB-enriched sympathetic neurons as regulators of adipose thermogenesis, revealing an unexpected neuron–ECM–adipocyte signaling pathway.
🔬 1. A specialized sympathetic neuron population
Single-cell RNA sequencing identified a neuronal subset enriched in β-glucuronidase (GUSB).
Approximately 25% of TH⁺ sympathetic neurons expressed GUSB.
Cold exposure selectively increased GUSB expression and neuronal activation.
⚡ 2. Sympathetic nerves remodel adipose ECM
Cold-activated neurons release GUSB into adipose tissue.
GUSB processes extracellular glycosaminoglycans, increasing bioavailable chondroitin sulfate (CS).
Cold → GUSB⁺ sympathetic neurons → Extracellular GUSB → ECM remodeling → CS release
Remarkably, this mechanism operates alongside classical norepinephrine signaling.
🧬 3. CS activates adipocyte thermogenesis through CD97
CS stimulates the adhesion GPCR CD97 (ADGRE5) on adipocytes.
CS → CD97 → PKA/CREB/HSL signaling → UCP1 ↑ → Thermogenic recruitment ↑
Adipocyte-specific CD97 deletion abolished CS-induced increases in oxygen consumption and thermogenic gene expression.
🔋 4. Genetic evidence establishes neuronal dependency
Sympathetic neuron-specific GUSB deletion reduced cold-induced UCP1 expression and oxygen consumption.
Recombinant GUSB or CS supplementation restored thermogenic responses.
In contrast, deleting GUSB from adipocytes, macrophages, fibroblasts, or endothelial cells did not reproduce this phenotype.
🔥 5. The pathway protects against diet-induced obesity
GUSB-deficient mice developed greater weight gain, glucose intolerance, insulin resistance, and impaired thermogenesis during high-fat feeding.
Intravenous CS administration (20 mg/kg every three days for eight weeks):
• Reduced weight gain and adiposity
• Improved glucose homeostasis
• Increased oxygen consumption
• Enhanced UCP1 expression
These benefits were substantially weakened by adipocyte CD97 deletion.
🧪 6. Human genetics suggests translational relevance
Obesity-associated GUSB promoter variants were identified.
KLF15 regulated GUSB transcription, whereas promoter variation disrupted KLF15-dependent activation in reporter assays.
However, human neuronal thermogenic mechanisms were not directly demonstrated.
🎯 Therapeutic implications
The GUSB–CS–CD97 axis represents a potential target for obesity and metabolic dysfunction.
Unlike conventional β-adrenergic approaches, it connects neural activity with extracellular matrix remodeling.
Nevertheless, evidence remains predominantly from male mice. Systemic CS may exert additional metabolic effects, and human efficacy and safety remain unestablished.
💡 Key takeaway: Adipose ECM is not merely structural scaffolding. Sympathetic neurons can enzymatically transform it into a source of bioactive signals that activate thermogenesis and influence whole-body metabolism.

What resonates most with me about this important paper by Jhodi Webster and Ashley Harms is that it challenges one of the assumptions that may be holding back progress in Parkinson’s disease: that we can treat a complex brain disease by targeting a single abnormal protein. The authors remind us that more than half of Parkinson’s brains examined after death harbor overlapping pathologies, including alpha-synuclein, tau, and amyloid-beta, and that these proteins may interact with one another and with the immune system to accelerate neurodegeneration and cognitive decline. I have long believed that Parkinson’s is not one disease, but many biological processes converging in the same person. What particularly caught my attention was the discussion of microglia, T cells, and chronic neuroinflammation, which may help explain why therapies targeting alpha-synuclein alone have repeatedly disappointed us. The lesson from cancer treatment is especially compelling: sometimes we need to attack several biological pathways simultaneously to meaningfully change the course of a disease. This does not mean combination therapies are proven to work in Parkinson’s, but it does suggest that our next generation of clinical trials should be smarter, more personalized, and designed around the biology of each person. We need to stop expecting one magic bullet to solve Parkinson’s. Our people (persons w/ disease) deserve treatments that recognize the full complexity of their disease.

Wow. Just look at those VLCFAs. They were among the first biomarkers to be obviously off for me when I first got sick. It was through learning about VLCFAs and peroxisomal dysfunction that I arrived at plasmalogen therapy as a potential treatment (Prodrome Plasmalogens were not even available back then). Since then metabolomics studies have been clear - lipids are dysfunctional in ME - phospholipids and plasmalogens are key if you want to protect your brain.
