New work connects #immunosenescence and the decline of the #thymus during aging with a drop in the ligand #RANKL—and shows RANKL treatment restores T cell production and rejuvenates immune responses in aged mice. @IrlaLab @JrmyCSantamaria @CIML_Immunology scim.ag/3ZAqihe
We provide the proof-of-concept that RANKL stimulates human endothelial and epithelial cells using thymic organo-cultures, providing an exciting new therapeutic approach in the arsenal of regenerative medicine to rejuvenate the immune system.
This increased production leads to peripheral T-cell renewal and improves T-cell-mediated anti-tumor and vaccine responses in aged mice.
RANKL treatment in aged mice restores thymic architecture as well as cellularity and functional properties of endothelial and epithelial cells. This boosted T-cell production in the aged thymus.
We demonstrate that the RANK-RANKL axis in the thymus is altered during aging. In addition to thymic epithelial cells, endothelial cells depend on RANK signaling for their cellularity and functional maturation.

One of the alluring and theoretical strategies for extending healthspan and longevity is to rejuvenate the thymus gland, promote an intact immune system. Now there's a way to do that in aged mice.

Vitamin D Deficiency in Childhood Linked to Higher Autoimmune Risk
A recent study has found that vitamin D deficiency during childhood accelerates the aging of the thymus, an organ essential for developing a healthy immune system.
The thymus helps train immune cells to distinguish between the body’s own tissues and harmful invaders, but when it ages prematurely due to a lack of vitamin D, it becomes less effective.
This increases the risk of autoimmune diseases like type 1 diabetes, as the immune system may mistakenly attack healthy tissues.
The research underscores the importance of adequate vitamin D intake, particularly during "vitamin D winter" when sunlight is insufficient for natural production.
Ensuring children receive proper vitamin D supplementation during these months could help prevent autoimmune disorders.
